It would be easier to spray them with the wheels off
End of the day either works. Both take almost the same amount of time unless you can get all four off at once ( do calipers at same time ). If you cant get all four off at once, garbage bag what you dont want done and remember to roll car back and forth to ensure all angles are done. You tube stuff by dipyour car.

Take them off and spray them. Lay the wheels down flat on the ground in the shade. Take your lug nuts and put them on the holes while you spray them. After each coat remove the lug nuts and repeat. This will prevent the tapered edge of the hole from getting paint on them. Because what will happen is when you put the wheels back on the car and tighten the lug nuts, they will grab onto the paint and twist it right off. Also when you tighten the nuts dont let the socket touch your wheels.
This happen to me recently. Had to remove the wheels, cut away what came off and respray around the holes.
Most people worry about the heat generated when braking which may cause the plastidip on the caliper to melt.. This could cause brake failure or a big mess lol. But let's be honest, we would have to be going 180 and coming to a complete screeching halt to reach such temperatures. You could try it on one and see how it fairs, but I'd say just use high temp/caliper paint to be safe.
